    Title: 大豆根瘤菌Sinorhizobium Fredii CCRC15769之聚半乳糖醛酸酶之生化特性

    Abstract: 聚半乳糖醛酸酶 (polygalacturonase, PG) 可能為根瘤菌與豆科植物根部進行共生固氮作用時,根瘤菌分解植物細胞壁的關鍵酵素。利用聚半乳糖醛酸(polygalacturonic acid) 當作基質,經PG作用五小時後,以BCA(2,2'-bicinchonic acid) 溶液測定反應液中顏色變化 (570nm) 定量酵素活性。本試驗以Sinorhizobium fredii CCRC 15769作為研究PG的菌株,嘗試以多種碳源誘導培養,並添加類黃酮誘導,以提高PG的產量。結果顯示,以myo-inositol為碳源可以得到較高的PG活性;而添加果膠、naringenin、DHF並不能對於PG活性有明顯的提升,genistein則具有提升活性的效果。菌體培養後經超音波破菌得到的粗酵素液,其作用最適pH為5.5,最適溫度為35℃。粗酵素液經硫酸銨劃分後,收集飽和濃度40-60%之沈澱,以20mM NaOAc緩衝液透析,離心後上清液通入Phenyl Sepharose 6之疏水性管柱,收取活性部分後透析,並繼續通入DEAE Spharose陰離子交換管柱,層析後收集具有活性的分劃,最後比活性(U/mg)為0.152,純化倍率為52.5,回收率為0.04%。
